The diagram shows the SITE100 workflow from field collecting to database entry. Samples from field plots are imaged and DNA is extracted from selected reference specimens, which are also used for shotgun sequencing (or long-range or multiple-short PCRs). The sequence database of (mostly) complete mitogenomes is used for tree construction. All other specimens are processed in bulk via metabarcoding, and after OTU clustering are subjected to phylogenetic placement. The growing database and tree are used for broader comparisons of global biodiversity.
